Compared with traditional culture methods, which take 24-72 hours to get results, flow cytometry can complete rapid quantitative detection of microorganisms within minutes, with a detection sensitivity that is 2-3 orders of magnitude higher than that of culture methods.
Flow cytometry employs hydrodynamic focusing technology, enabling microorganisms in water samples to form a microfluidic single-cell fluid encapsulated by sheath fluid, which then rapidly passes through the laser detection zone. Through specific fluorescent staining and scattered light signal analysis, the system can obtain information such as cell viability, DNA content, cell size, and structural complexity. Flow cytometry is capable of distinguishing between intact and damaged bacteria, and can detect "viable but non-culturable (VBNC)" microorganisms that are unidentifiable by traditional culture methods.

In traditional culture-based testing, microorganisms are counted manually during the final colony formation stage, which is time-consuming.
